I am new to Toyota. I have read various threads that claim Toyota understates clearance specs. I believe factory spec on my 27 LC250 is 8.6". I have Falken 33's on the vehicle now. Any opinion on what my "real" clearance might be?

I’ve heard folks on YouTube say the same thing. But I don’t have an answer for you.
I have same 33” AT4W + Westcott 2.5”F/1”R lift, & the lift isn’t applied @ 1:1 in lift over stock. But what it did do is make the bottom of the rear diff the lowest point, where prior in stock form the middle of the front edge of the skip plate was lowest point. So leveling has merit. Using a tape our LC is @ 10.25” @ diff (lowest), & 10.5” @ front skid. If concerned, makes all the more reason to add the SLEE reinforcement for the bottom of the radiator core support.
This is really it, OP. It’s a really good question you ask, and the absolute best way to answer it is also super easy: just grab a tape and go outside. I bet you’ll find the front skid plate to be the true lowest point if you have one, not the rear diff, but I might be wrong. Let us know what you measure!
Yep and you also improve the approach angle a bit with the leveling kit.
